What Is Gram Stain?

Gram Stain is a type of staining technique used to identify bacteria by detecting proteins and cell walls. The technique is based on the differences of the cell walls of bacteria, which allows the bacteria’s genus to be determined.

Preparation for a Gram Stain

To perform a Gram stain, you will need a slide with a bacterial sample, crystal violet dye, iodine solution, ethanol, safranin, and a microscope. The slide must be prepared with a bacterial sample, which is usually obtained by scraping a wound or a culture that contains the bacteria.

The Procedure for a Gram Stain

The Gram Stain technique is performed using the following process:

  • Apply crystal violet to the bacterial sample
  • Rinse off the crystal violet with water
  • Apply iodine solution to the sample
  • Rinse off the iodine solution with water
  • Apply ethanol to the sample
  • Rinse off the ethanol with water
  • Apply safranin to the sample
  • Rinse off the safranin with water
  • Observe the sample under the microscope to determine the bacteria’s genus

Types of Bacteria According to the Gram Stain

The Gram Stain technique allows for bacteria to be categorized into two groups: Gram-positive and Gram-negative. Gram-positive bacteria are characterized by their thick cell wall, which prevents the crystal violet from being washed away. Gram-negative bacteria have a thin cell wall, which allows the crystal violet to be washed away.

Risks Associated with Gram Staining

There are no major risks associated with performing a Gram Stain. However, the dye used in the technique may be hazardous if it comes in contact with the eyes or skin. It is important to wear eye protection when performing a Gram Stain.
