Mirolast binds to the histamine H1 receptor. This blocks the action of endogenous histamine, which subsequently leads to temporary relief of the negative symptoms brought on by histamine. Mirolast has also been observed to inhibit antigen-stimulated calcium ion influx into mast cells through the blockage of calcium channels. Mirolast inhibits the chemotaxis of eosinophils into ocular tissue, and prevents inflammatory mediator release from human eosinophils.

Mirolast is used for the prophylactic treatment of itching of the eye associated with allergic conjunctivitis. Mirolast potassium is a mast cell stabilizer that inhibits the in vivo Type I immediate hypersensitivity reaction. Mirolast inhibits the antigen-induced release of inflammatory mediators (e.g., histamine, leukotriene C4, D4, E4) from human mast cells. Allergic reactions lead to cell-degranulation and the release of histamine (and other chemical mediators) from the mast cell or basophil. Once released, histamine can react with local or widespread tissues through histamine receptors. Histamine, acting on H1-receptors, produces pruritis and vasodilatation (allowing blood fluids to enter the area to cause swelling). Mirolast is a histamine H1 antagonist. It competes with histamine for the normal H1-receptor sites on effector cells of blood vessels to provide effective, temporary relief of watery and itchy eyes.

Dosage

Mirolast dosage

Adult: 1 to 2 drops in each affected eye four times daily.

Pediatric Use: Safety and effectiveness in pediatric patients below the age of 3 years have not been established.

Side Effects

Burning of eye, dry eye, ocular discomfort, non ocular allergy, sinusitis, headache and sneezing or nasal congestion.

Precaution

To prevent contaminating the dropper tip and solution, do not touch the eyelids or surrounding areas with the dropper tip. Keep the bottle tightly closed when not in use Patients should be advised not to wear contact lens if their eye is red Mirolast should not be used to treat contact lens related irritation Patients should wait 10 minutes after applying Mirolast before they insert contact lenses

Elimination Route

Following topical administration, about 10-15% of the dose was excreted unchanged in the urine.

Pregnancy & Breastfeeding use

Pregnancy: There are no adequate and well-controlled studies in pregnant women. Mirolast ophthalmic solution should be used during pregnancy only if the benefit outweighs the risk.

Lactation: It is not known whether Mirolast Potassium is excreted in human milk. Because many drugs are excreted in human milk, caution should be exercised when Mirolast ophthalmic solution is administered to a nursing woman.

Contraindication

Mirolast ophthalmic solution is contraindicated in patients with previously demonstrated hypersensitivity to any of the ingredients of this product.

Storage Condition

Store at 15-25° C. Keep out of reach of children. Do not use more than 4 weeks after opening
